  OBJECTIVE: Estimation of protein by the Biuret method (Qualitative Analysis)  REQUIREMENTS:  Test tubes  Measuring cylinder  Dropper Reagents: Protein Sample  CuSO4 NaOH  THEORY:  This is the most commonly used method based on the fact that the –CO – NH- (peptide) group of proteins forms a purple complex with copper ions in an alkaline medium. Since all proteins contain the peptide bond. The method is fairly specific and there is little interference from other compounds. The normal color of biuret reagent is blue. The reagent turns violet in the presence of peptide bonds. The chemical bonds that hold amino acids together.
